This is a working overview of regulatory approval, written for readers who want more than a one-paragraph summary but less than a textbook.
Reviewed 2026-03-15. Anything still debated is marked as such rather than presented as settled.
Dissolution is usually performed in water or a suitable aqueous buffer, and the resulting liquid is divided into single-use portions before freezing. Freeze-thaw cycling is a recognised source of loss for short peptides, since each cycle can encourage aggregation or adsorption onto container walls. Working solutions are generally kept cold and used within a short window, although published stability data specific to epitalon are thin. Containers should be marked with concentration and date, and solutions examined for cloudiness before use.
Identity is normally established by reversed-phase high-performance liquid chromatography combined with mass spectrometry, a pairing that separates components and confirms molecular mass at once. Purity is quoted as a percentage from the chromatogram, and figures above ninety-five percent are a frequent commercial specification. Amino acid analysis or sequencing supplies further confirmation when required. Because many short peptides behave similarly under chromatography, retention time alone does not establish sequence; the mass measurement is what separates one tetrapeptide from another, and certificates should report both.
Short peptides such as AEDG are normally supplied as a freeze-dried solid and are kept dry, cold, and shielded from light. Holding at minus twenty degrees Celsius is common for the long term, while a refrigerator suffices for brief periods before use. The material takes up moisture to some degree, so containers should stay sealed and be allowed to reach room temperature before opening, which limits condensation on the contents. Repeated warming and cooling of a single container is discouraged because it admits water and can lower the amount of intact peptide.
Literature searches for this compound must account for several spelling variants. Indexing databases contain epitalon, epithalon, epithalone, and AEDG, and relevant records are scattered across Russian-language and English-language journals that do not consistently cross-cite. Early publications describe the parent extract as a mixture of many peptides, whereas later work addresses the single synthetic tetrapeptide. That shift in nomenclature complicates comparison between studies, because extract data and tetrapeptide data are sometimes cited interchangeably. A search strategy omitting the alternate spellings will return an incomplete set of references.
Reported biological findings come mainly from cell culture and rodent experiments. Those studies describe changes in telomerase catalytic subunit expression, melatonin rhythm amplitude, and antioxidant enzyme activity after peptide exposure. Human data are sparse and consist of small trials with limited blinding and inconsistent endpoints. The proposed mechanisms remain hypotheses rather than established facts, and there is no consensus on whether effects observed in animals carry over to people. Independent replication outside the original research groups is limited, which is a recognised gap in the published literature.

No national medicines regulator has approved epitalon as a therapeutic product. It is generally distributed as a research chemical, and in some jurisdictions selling peptides for human consumption without approval is restricted or prohibited. Certificates of analysis accompanying commercial material vary in which tests are performed, and independent verification of identity and purity is uncommon. Statements about anti-ageing or disease-prevention benefits on vendor pages are marketing claims rather than regulatory findings, a distinction that shapes how the compound is discussed in scientific and popular sources alike.
The most frequently cited laboratory finding is that AEDG increased telomerase activity and extended telomere length in cultured human somatic cells. That work used fetal fibroblast strains and reported changes in enzyme activity alongside altered division counts. Replication by unrelated groups has been limited, and the published record is largely a single-laboratory series rather than a multi-centre programme. The result supports a hypothesis about peptide influence on gene expression in cell culture; it does not by itself establish an effect on telomere length in living animals or in people.
The compound is described in the literature as a derivative of epithalamin, a preparation obtained from bovine pineal tissue. Work on this peptide family was carried out mainly by a research group in Saint Petersburg beginning in the 1980s, and the substance was later registered for clinical use in Russia under the name Epitalon. Outside that region it is generally treated as a research chemical rather than an approved medicine. Statements about its biological activity rest on a relatively small number of studies, and independent replication remains limited.
In its supplied form epitalon is a white to off-white powder, usually lyophilized and often hygroscopic. It dissolves readily in water and in aqueous buffers, and it is commonly handled as the acetate or trifluoroacetate salt to improve stability during purification and drying. The amide backbone is labile in aqueous media, so solutions are less durable than the dry solid. Handling notes in the chemical literature therefore emphasize keeping the powder dry, cool and shielded from light until it is dissolved.
Epitalon is a synthetic tetrapeptide whose sequence is alanine-glutamate-aspartate-glycine, written in single-letter code as AEDG. The four residues are joined by three peptide bonds, giving a linear backbone with no branching and no disulfide bridges. Its calculated molecular mass for the free form is approximately 390.3 daltons, a figure that rises when the compound is supplied as an acetate or trifluoroacetate salt. Because the chain is short, the molecule is defined entirely by its residue order rather than by any folded three-dimensional structure.
Peptides of this size are generally stable as dry solids but degrade in solution over time. The principal routes are hydrolysis of the peptide backbone and oxidation, with hydrolysis favoured by elevated temperature and extreme pH. Aqueous solutions held at room temperature can show measurable loss of purity within days, while frozen aliquots are considerably more durable. Because the sequence contains neither cysteine nor methionine, oxidation is less of a concern than for many other peptides, but pH control during handling still matters.
Typical storage for the lyophilized powder is −20 °C or lower, in a sealed container protected from light and moisture. Hygroscopic material should be allowed to equilibrate to room temperature before the vial is opened, which limits condensation on the contents. Working solutions are commonly divided into single-use aliquots and frozen to avoid repeated freeze-thaw cycles. Dilute solutions are more prone to adsorption onto plastic surfaces and to loss during filtration, so procedures that minimize transfers and use low-binding labware are preferable.
Epitalon is a synthetic tetrapeptide with the sequence alanine-glutamate-aspartate-glycine, abbreviated AEDG. Its molecular formula is C14H22N4O9 and its calculated monoisotopic mass is approximately 390.35 daltons. The compound does not occur naturally as a free peptide; it is produced by solid-phase peptide synthesis. Because it contains two acidic residues and no basic residues, the neutral form carries a net negative charge at physiological pH. This charge profile influences how the peptide behaves in solution and during chromatographic analysis.
Laboratory-grade epitalon is typically supplied as a lyophilized powder. Purity is commonly assessed with reverse-phase high-performance liquid chromatography, often paired with mass spectrometry to confirm molecular identity. Amino acid analysis and peptide mapping can provide additional confirmation of sequence. Certificates of analysis for research materials frequently report purity above 95 percent, although the methods behind such figures vary between suppliers. The absence of a pharmacopeial monograph means that no single standardized assay defines the compound, so reported results depend on the analytical protocol chosen.
Storage recommendations center on limiting moisture, heat, and light. The dry powder is generally kept at minus 20 degrees Celsius, and some suppliers recommend minus 80 degrees for long-term archival. Once dissolved, solutions are usually aliquoted and frozen to avoid repeated freeze-thaw cycles, which can promote aggregation or degradation. Aqueous stability depends on pH and concentration, and buffered saline is often preferred over plain water for biological work. Stability data specific to epitalon remain limited, so general peptide-handling practices are applied by analogy rather than from product-specific validation.
==== Protease inhibitors ==== Some viruses include an enzyme known as a protease that cuts viral protein chains apart so they can be assembled into their final configuration. HIV includes a protease, and so considerable research has been performed to find "protease inhibitors" to attack HIV at that phase of its life cycle. Protease inhibitors became available in the 1990s and have proven effective, though they can have unusual side effects, for example causing fat to build up in unusual places. Improved protease inhibitors are now in development. Protease inhibitors have also been seen in nature. A protease inhibitor was isolated from the shiitake mushroom (Lentinus edodes). The presence of this may explain the Shiitake mushrooms' noted antiviral activity in vitro.
== Disease cycle == Pseudomonas syringae overwinters on infected plant tissues such as regions of necrosis or gummosis (sap oozing from wounds on the tree) but can also overwinter in healthy looking plant tissues. In the spring, water from rain or other sources will wash the bacteria onto leaves/blossoms where it will grow and survive throughout the summer. This is the epiphyte phase of P. syringae's life cycle where it will multiply and spread but will not cause a disease. Once it enters the plant through a leaf's stomata or necrotic spots on either leaves or woody tissue then the disease will start. The pathogen will then exploit and grow in intercellular space causing the leaf spots and cankers. P. syringae can also survive in temperatures slightly below freezing. These below freezing temperatures increase the severity of infection within trees like sour cherry, apricot, and peach.
=== Analogues === The closely related medication, methylnaltrexone (N-methylnaltrexone), is used to treat opioid-induced constipation but does not treat addiction as it does not cross the blood–brain barrier. Nalmefene (6-desoxy-6-methylenenaltrexone) is similar to naltrexone and is used for the same purposes as naltrexone. Naltrexone should not be confused with naloxone (N-allylnoroxymorphone), which is used in emergency cases of opioid overdose. Other opioid antagonists related to naltrexone include 6β-naltrexol (6β-hydroxynaltrexone), samidorphan (3-carboxamido-4-hydroxynaltrexone), β-funaltrexamine (naltrexone fumarate methyl ester), nalodeine (N-allylnorcodeine), nalorphine (N-allylnormorphine), and nalbuphine (N-cyclobutylmethyl-14-hydroxydihydronormorphine).
The Solomon Islands-China security pact caused alarm in late 2022, as China could build military bases across the South Pacific. Biden sought to strengthen ties with Australia and New Zealand in the wake of the deal. In a September 2022 interview with 60 Minutes, Biden said that U.S. forces would defend Taiwan in the event of "an unprecedented attack" by the Chinese, which is in contrast to the long-standing U.S. policy of "strategic ambiguity" toward China and Taiwan. The September comments came after three previous comments by Biden that the U.S. would defend Taiwan in the event of a Chinese invasion. Amid increasing tension with China, Biden's administration has repeatedly walked back his statements and asserted that U.S. policy toward Taiwan has not changed. In late 2022, Biden issued several executive orders and federal rules designed to slow Chinese technological growth, and maintain U.S. leadership over computing, biotech, and clean energy. On February 4, 2023, Biden ordered the United States Air Force to shoot down a suspected Chinese surveillance balloon off the coast of South Carolina. The Chinese government denied that the balloon was a surveillance device, instead claiming it was a civilian airship that had blown off course. Secretary of State Antony Blinken postponed his planned visit to China as the incident further damaged U.S.-China relations. In May 2024, the Biden administration doubled tariffs on solar cells imported from China and more than tripled tariffs on lithium-ion electric vehicle batteries imported from China.
In children they may be used in those with disruptive behavior disorders, mood disorders and pervasive developmental disorders or intellectual disability. Antipsychotics are only weakly recommended for Tourette syndrome, because although they are effective, side effects are common. The situation is similar for those on the autism spectrum. Much of the evidence for the off-label use of antipsychotics (for example, for dementia, OCD, PTSD, personality disorders, Tourette's) was of insufficient scientific quality to support such use, especially as there was strong evidence of increased risks of stroke, tremors, significant weight gain, sedation, and gastrointestinal problems. A UK review of unlicensed usage in children and adolescents reported a similar mixture of findings and concerns. A survey of children with pervasive developmental disorder found that 16.5% were taking an antipsychotic drug, most commonly for irritability, aggression, and agitation. Both risperidone and aripiprazole have been approved by the US FDA for the treatment of irritability in autistic children and adolescents. A review in the UK found that the use of antipsychotics in England doubled between 2000 and 2019. Children were prescribed antipsychotics for conditions for which there is no approval, such as autism. Aggressive challenging behavior in adults with intellectual disability is often treated with antipsychotic drugs despite lack of an evidence base.

=== Organic chemistry research === Oleum is a harsh reagent, and is highly corrosive. One important use of oleum as a reagent is the secondary nitration of nitrobenzene. The first nitration can occur with nitric acid in sulfuric acid, but this deactivates the ring towards further electrophilic substitution. A stronger reagent, oleum, is needed to introduce the second nitro group onto the aromatic ring.
Defending against a full Russian or Chinese attack would require hundreds of thousands of satellites," according to MIT physicists Lisbeth Gronlund and David Wright. The cost of launching satellites is expected to decrease when the heavy-lift SpaceX Starship launch vehicle enters service. Still, the extreme expense of creating such a constellation—and replacing satellites as their orbits decay—has led the U.S. military to also pursue space-based midcourse options that would require fewer satellites. But these would be more expensive and no better than ground-based interceptors at telling decoys from warheads. Space-based interceptors would be guided by the nascent Hypersonic and Ballistic Tracking Space Sensor system, one of the tracking elements of the Space Development Agency's (SDA) seven-layer Proliferated Warfighter Space Architecture (PWSA). The Golden Dome would also be aimed at detecting and countering strikes delivered from a fractional orbital bombardment system (FOBS). Pentagon leaders have said the system could be used to put offensive weapons in space, giving a global strike capability that would create offensive overmatch. Defense Secretary Pete Hegseth has said the U.S. "will be able to strike anywhere, anytime". Satellites might carry the bunker-busting Common-Hypersonic Glide Body (C-HGB), which can reportedly fly 1,000 miles to strike within 14 inches of its target. The missile was designed to fit the HIMARS ground launcher, so several can be compactly racked in a satellite payload bays.
Friedrich Wöhler discovered that an organic substance, urea, could be produced from inorganic starting materials in 1828. That was an important conceptual milestone in chemistry by being the first example of a synthesis of a substance that had been known only as a byproduct of living processes. Wöhler obtained urea by treating silver cyanate with ammonium chloride, a simple, one-step synthesis:
There are many different kinds of antibody and their production is highly complex, involving several types of lymphocyte, but in general lymphocytes and other antibody regulating and producing cells will migrate to the lymph nodes to aid in the release of these antibodies into the bloodstream. On the other hand, there are natural killer cells; these cells have the ability to take down threats that are larger in size, such as bacteria, parasites, and tumor cells. A separate study found that cortisol effectively disarmed natural killer cells, downregulating the expression of their natural cytotoxicity receptors. Prolactin has the opposite effect. It increases the expression of cytotoxicity receptors on natural killer cells, increasing their potency. Cortisol stimulates many copper enzymes (often to 50% of their total potential), including lysyl oxidase, an enzyme that cross-links collagen and elastin. Especially valuable for immune response is cortisol's stimulation of the superoxide dismutase, since this copper enzyme is almost certainly used by the body to permit superoxides to poison bacteria. Some viruses, such as influenza and SARS-CoV-1 and SARS-CoV-2, are known to suppress the secretion of stress hormones to avoid the organism's immune response. These viruses suppress cortisol by producing a protein that mimics the human ACTH hormone but is incomplete and does not have hormonal activity. ACTH is a hormone that stimulates the adrenal gland to produce cortisol and other steroid hormones.
== Function == This gene encodes the alpha chain of type XVI collagen, a member of the FACIT collagen family (fibril-associated collagens with interrupted helices). Members of this collagen family are found in association with fibril-forming collagens such as type I and II, and serve to maintain the integrity of the extracellular matrix. High levels of type XVI collagen have been found in fibroblasts and keratinocytes, and in smooth muscle and amnion.

=== Biomedical === Because one of the main purposes for biomedical engineering is to mimic body parts to sustain normal body functions, due to their biocompatible properties, biopolymers are used vastly for tissue engineering, medical devices and the pharmaceutical industry. Many biopolymers can be used for regenerative medicine, tissue engineering, drug delivery, and overall medical applications due to their mechanical properties. They provide characteristics like wound healing, and catalysis of bioactivity, and non-toxicity. Compared to synthetic polymers, which can present various disadvantages like immunogenic rejection and toxicity after degradation, many biopolymers are normally better with bodily integration as they also possess more complex structures, similar to the human body. More specifically, polypeptides like collagen and silk, are biocompatible materials that are being used in ground-breaking research, as these are inexpensive and easily attainable materials. Gelatin polymer is often used on dressing wounds where it acts as an adhesive. Scaffolds and films with gelatin allow for the scaffolds to hold drugs and other nutrients that can be used to supply to a wound for healing. As collagen is one of the more popular biopolymers used in biomedical science, here are some examples of their use: Collagen based drug delivery systems: collagen films act like a barrier membrane and are used to treat tissue infections like infected corneal tissue or liver cancer. Collagen films have all been used for gene delivery carriers which can promote bone formation.
Fibrillogenesis is the expansion of fine fibrils which is common in collagen fibers of connective tissue. The definite mechanisms of fibrillogenesis are still unknown, although many hypotheses resulting from basic research help discover many possible mechanisms. In early experiments, collagen I could be distilled from tissues and recombined into fibrils with controlling the solutions. Later studies help understand the composition and structure of binding sites on the collagen monomers. Collagen is synthesized as a soluble precursor, procollagen, which supports collagen self-assembly. Since collagen fibrils have almost 50 binding components in vivo, the definite requirement to generate fibrillogenesis in vivo is still cryptic. With acidic or saline solution, collagen can be extracted from tissues and rearrange into fibril by changing temperature or pH value. Experiments discovered attractive force between collagen monomers which helps the rearrangement. Collagen serves as a precursor, procollagen, in synthesizing reaction, which identifies self-polymerization of collagen.
=== Searches for element 61 === In 1902, Czech chemist Bohuslav Brauner found out that the differences in properties between neodymium and samarium were the largest between any two consecutive lanthanides in the sequence then known; as a conclusion, he suggested there was an element with intermediate properties between them. This prediction was supported in 1914 by Henry Moseley who, having discovered that atomic number was an experimentally measurable property of elements, found that a few atomic numbers had no known corresponding elements: the gaps were 43, 61, 72, 75, 85, and 87. With the knowledge of a gap in the periodic table several groups started to search for the predicted element among other rare earths in the natural environment. The first claim of a discovery was published by Luigi Rolla and Lorenzo Fernandes of Florence, Italy. After separating a mixture of a few rare earth elements nitrate concentrate from the Brazilian mineral monazite by fractionated crystallization, they yielded a solution containing mostly samarium. This solution gave x-ray spectra attributed to samarium and element 61. In honor of their city, they named element 61 "florentium". The results were published in 1926, but the scientists claimed that the experiments were done in 1924. Also in 1926, a group of scientists from the University of Illinois at Urbana–Champaign, Smith Hopkins and Len Yntema published the discovery of element 61. They named it "illinium", after the university.
=== Violent Crime and Law Enforcement Act === Clinton signed the 1994 Crime Bill, which was written by Senator Joe Biden of Delaware. It included 30 billion dollars' worth of federal anticrime funding. Specific to the war on drugs, Clinton included the three-strikes law, which required a mandatory life sentence for any felony violent crime conviction after two other prior convictions. Some social justice critics argue that the law disproportionately affected criminals in urban neighborhoods hurt by policing the war on drugs, rather than white-collar criminals. The bill also authorized $16 million in new funding for the development of federal prisons and state and local police forces. The Justice Policy Institute stated in 2008 that the administration's "tough on crime" policies caused larger increases in federal and state inmate populations than for any other president in American history. The act also saw the expansion of the federal death penalty. Sixty new capital punishment charges were created including non-homicidal narcotics offenses, drive-by shootings resulting in death, and carjackings relating to death. Between 1994 and 1999, nearly two thirds of people sentenced to the federal death penalty were black during Clinton's presidency, nearly seven times that of their representation in the US population. During his two terms, the federal prison population increased from 1.3 million to 2 million inmates. The law featured a provision that removed the approval of Pell Grants for low-income inmates in federal prison.

Reversed-phase high-performance liquid chromatography is run alongside mass spectrometry. The chromatogram separates components and gives a purity figure, while the mass spectrum confirms that the measured molecular mass matches the expected sequence. Neither measurement on its own is treated as sufficient.
Long-term holding is usually at minus twenty degrees Celsius in a sealed, desiccated container. A refrigerator is adequate for short intervals before use. Light exposure is normally avoided as well. Allowing a cold container to warm before opening reduces moisture condensation on the contents.
Repeated cycles are generally avoided. Each freeze and thaw can promote aggregation or loss of peptide to container surfaces, which reduces the amount available for later work. Dividing a solution into single-use portions at the outset is the usual way to limit this problem.
It is based on epithalamin, a peptide-rich preparation obtained from pineal gland tissue and studied in the former Soviet Union. Researchers later identified the tetrapeptide AEDG as a constituent fragment of that material. Modern supplies are produced by chemical synthesis rather than tissue extraction.
